The Danish frontman will start for the first time in a black and white shirt in the third round tie on Tyneside as Magpies head coach Eddie Howe rings the changes for the visit of the League Two side.
Howe has made eight alterations to his side following Saturday's 1-1 draw with Manchester City in the Premier League, with only Joelinton, Harvey Barnes and Fabian Schär - who captains United - remaining in the line-up from the weekend clash with the champions.
Martin Dúbravka starts in goal, with Tino Livramento, Emil Krafth, Schär and Lloyd Kelly at the back and Joe Willock, Sean Longstaff and Joelinton in midfield.
And up front, forward Osula will have support from Barnes and Miguel Almirón on the flanks.
There are two goalkeepers - Odysseas Vlachodimos and John Ruddy - on the bench, along with defenders Dan Burn, Alex Murphy and Lewis Hall, while Sandro Tonali and Bruno Guimarães are also named.
Jacob Murphy and Anthony Gordon represent attacking reinforcements on the bench for Newcastle, who are looking for the win that would book a visit from Chelsea in the fourth round.
