Cheatley's two goals took Brentford into half-time with a comfortable lead before Chloe Logie struck moments after the break to make it 3-0 to the hosts.
Ascot managed to claw their way back into the game with two goals in less than two minutes in what was a frantic start to the second half.
The home side did well to weather the Ascot storm as they went on to win the game with Logie netting her second in stoppage time as the Bees secured an historic win.
Brentford headed into the first-round proper in incredible form, scoring 32 goals in the previous three rounds of the competition, winning 18-1 win against Northleigh, 12-1 against Tilehurst Panthers and 2-0 against Sport London E Benfica.
Cheatley’s two goals against Ascot took her to nine goals so far in the competition.
Brentford continued their incredible run of form which has seen the side win every game they have played so far this season.
The home side put on a brilliant display in front of 500 fans, beating Ascot who play their league football in the division above.

Brentford faced London Seaward most recently in a pre-season fixture in August - the Bees came out 5-3 winners.
